胶原的类型有很多，目前被确认的有四十多种，常见类型为Ⅰ型、Ⅱ型、Ⅲ型、Ⅴ型和Ⅺ型，存在于人体的皮肤，软骨，肌腱等组织内，占人体或者其他动物总蛋白量的25%-33%。胶原蛋白的定量检测对于某些疾病的诊断尤为重要，今天给大家带来几款艾美捷代理品牌Biocolor公司的专利产品，超过1000篇文献验证，Nature，PNAS，ACS Nano这些顶级期刊都在用的胶原蛋白检测试剂盒。
1、Sircol Soluble Collagen Assay（可溶性胶原检测试剂盒，货号：S1000）
该试剂盒采用与胶原特异性结合的Sircol染料，主要用于定量检测哺乳动物组织中抽提的酸溶性胶原和体外培养哺乳动物细胞释放到培养基中的胶原。
可溶于酸、胃蛋白等的可溶性胶原均可检测，具体可检测的胶原类型为I-V型，VI-XIV型胶原也可检测，但由于难以制备标准曲线，还无法定量。Sircol染料不能区分不同类型的胶原，但可与胶原的[Gly-X-Y]n螺旋结构特异性地结合。随着胶原变性为凝胶，该染料与胶原的结合力减弱。非脊椎动物与胶原由于变性温度较低、羟脯氨酸残基较少等原因，结合染料的能力较弱，需要制备种属特异性的纯化标准品，以绘制标准曲线。
2、Sircol Insoluble Collagen Assay（不可溶性胶原检测试剂盒，货号：S2000）
这种检测方法能用于评估因快速生长，发育，组织修复，重塑和创伤愈合而新近沉积的胶原纤维的产生速度。适用的样本包括组织，骨骼和钙化的组织。
特点：
- 它替代了常规羟脯氨酸分析方法。无需操作浓酸和浓碱。
- Sircol不溶性胶原检测方法使用温和的酸和温度处理，耗时2到3小时。
- 经溶解性转换为变性胶原，后续检测在一天内完成。
